Melting temperature (Tm)
The temperature at which half of a duplex has separated into single strands.
The melting temperature, or Tm, is the temperature at which half of a double stranded oligonucleotide has separated into single strands. It reflects how tightly the two strands bind, which depends on length, GC content, sequence and salt concentration.
Tm is a key design parameter for primers, probes and antisense oligos, and can be estimated with the Syngenis oligo properties calculator.
